Martha's parents were Francis Littleberry Eppes III and Anne Isham. Both the Eppes and Isham families were of English heritage.
Her paternal great-grandfather Col. Francis Eppes I was born in May 1597 in Ashford, Kent, England. Another date of his birthday which I have come across is 15 May 1588.

Francis Eppes I emigrated to Virginia around 1620, he died in Charles City.
Martha's maternal grandfather was Henry Isham I, born around 1627 in Northamptonshire, England. In the year 1656, he came to America and started his new life in Bermuda Hundred, Henrico County, Virginia Colony.
In 1688, Martha's great-grandfather Francis and her uncle (by marriage with Mary Isham, Martha's mother's sister) William Randolph set out the Bermuda Hundred town in Virginia.
Martha Eppes was also related to President Thomas Jefferson in two ways:

There was another family connection between the Epps and the Jeffersons - Francis Eppes VI's wife Mrs. Elizabeth Eppes nee Wayles (b. 24 Feb. 1752, VA / d. 10 June 1810, Eppington Plantation, Chesterfield, Virginia) was a half-sister of Martha Jefferson nee Wayles (b. 30 Oct. 1748, Charles City, VA / d. 6 Sept. 1782, Monticello, Charlottesville, VA), wife of Thomas Jefferson.
